OBJECT RECOGNITION

This procedure is based on the methods described in: Dodart, J.-C., Meziane, H., Mathis, C., Bales, K. R., Paul, S. M., & Ungerer, A. (1999) Behavioral disturbances in transgenic mice overexpressing the V717F b-amyloid precursor protein. Behavioral Neuroscience, 113:982-990.

Apparatus: 95% ETOH, C57Bl6/H mice, 10 gallon aquarium spray painted black, clear Plexiglas cage top, "objects": glass marbles, dice, bolts glued to Plexiglas, timer with at least two stop watches

Procedure:

Beforehand
1. Cut out and place velcro on bottom of Plexiglas plate below each object. (Check which type of velcro is already in the aquarium corners to determine which type of velcro to put on bottom)

Habituation (Day 1)
1. Attach bolts to aquarium floor (should be attached on bottom by velcro).
2. Turn off over head lights and trn on 60W lamp on computer table.
3. Get timer ready.
4. Place mouse in aquarium and allow it to explore for 30 min.
5. Remove mouse and wash aquarium with 95% ETOH. Also wipe off objects with ETOH or simply immerse them in a cup of ETOH.

Testing (Day 2)
1. Familiarization

a. Put two of the same objects (either two dice or two glass marbles) into aquarium.
b. Place mouse into aquarium.
c. Allow mouse to explore for ten minutes. Record time spent exploring each of the two objects.
d. Remove mouse and wash aquarium with 95% ETOH. Also wipe off objects with ETOH or simply immerse them in a cup of ETOH.
2. Delay
a. Wait for two hours.
b. Return mouse to home cage during delay.
3. Test
a. Place one familiar object and one novel object into aquarium.
b. Place mouse back into aquarium.
c. Allow mouse to explore for 10 minutes.
d. Record time spent at each object.
e. Remove mouse and wash aquarium with 95% ETOH. Also wipe off objects with ETOH or simply immerse them in a cup of ETOH.


Scoring
1. Exploration is exploration, not loitering. Score an animal as exploring (i.e. have timer going) when:

a. Mouseís nose is within 1 cm of object and vibrissa are moving (i.e. sniffing).
b. If vibrissa are not moving and mouse is frozen, donít score.
c. If mouse is standing on odor cup to get a better view of something else, donít score.
d. If mouse is within 1 cm of odor cup but pointed away from cup, donít score.
