ROTOROD

Apparatus: Economex Rotorod from Columbus Instruments, mice, data sheet for recording latency to fall times

Procedure:
1. HANDLE ALL MICE DURING TESTING BY BASE OF TAIL!!!

Stationary training (non-moving rod and constant 2.5 rpm rate)
1. Bring mice to testing room.
2. Check settings on rotorod (RR). The rpm should be set at 1.0 (really 2.5 rpm), the accelerator should be set at 0.0 (allows for acceleration of 2.5 20 rpm from 0 180 seconds), sensitivity should be set at 7.0.
3. Plug unit in at beginning of testing and unplug at end of day if surge strip is not available.
4. The rod may not rotate at these settings on its own turn the rpm up to 2.0 2.5 and then back down to 1.0 rpm before each test.
5. Training on stationary rod.

a. Place mouse on rod facing back wall of RR.
b. Start timer at bottom of rod for that mouse.
c. Allow mouse to stay on rod until it falls or 60 sec have elapsed; record latency to fall.
d. Repeat this sequence 2 more times (three trials total) with no intertrial interval.
e. If mouse stays on rod for a total of 60 seconds over the three trials then go onto step 6.
f. If mouse does not stay on rod for total of 60 seconds, repeat step 5 procedure (three trials in a row) once more on day 1 approximately 20 minutes after the end of the first session.
g. Continue training on stationary rod for two sessions (three trials each session) a day (with a 20 minutes intersession interval) until criterion of 60 seconds for one session (three trials) has been met, and then go to step 6.
6. Training on constant 2.5 rpm rate.
a. Turn on RR, but DO NOT press accelerating button. Rod should be moving at 2.5 rpm.
b. Place mouse on moving rod facing back wall.
c. Allow mouse to stay on rod until it falls or 60 seconds have elapsed; record latency to fall.
d. Repeat last two steps 2 more times (three trials total) with no intertrial interval.
e. If mouse stays on rod for a total of 60 seconds over the three trials then go onto accelerating test.
f. If mouse does not stay on rod for total of 60 seconds, repeat step B.2.f. (three trials in a row) once more on day 1 approximately 20 minutes after the end of the first session.
g. Continue training on constant 2.5 rpm rate rod for two sessions (3 trials each session) a day (with a 20 min intersession interval) until criterion of 60 seconds for one session (three trials) has been met, and then go to accelerating test .
Accelerating test (2.5 20 rpm over 180 seconds)
1. Turn on rod at 2.5 rpm rate.
2. Place mice on rod facing back wall.
3. Some mice may fall off while others are being placed on the rod. I try to put the fallen mice back on the rod a total of 2 more times. If they can not stay on the rod for 2 more times at the 2.5 rpm rate, I give them a score of 0 seconds.
4. Once all mice have been placed on rod, press accelerating button. Record latency to fall.
5. If mouse stays on rod for 180 seconds, then stop trial and record 180 seconds for that mouse.
6. Repeat steps 2-5 two more times with an average intertrial interval of 10 minutes.
7. Total of three trials per day.
8. Test for a total of five days.
